Stable isotope analysis of human remains is a research tool that can provide paleodiet information for archaeological sites, such as caves, where traditional evidence may be missing or out of context. Unlike other lines of evidence, the stable isotopes of carbon and nitrogen in human bone reflect the chemistry of the diet and therefore provide a direct measure of the foods consumed. As an example, the data from isotopic analyses of bone from the Mer site (44LE280), a cave in Lee County, Virginia, are presented. Although this site lacks faunal and floral remains to provide basic information about the availability and potential utilization of food resources, the stable isotope data from other sites throughout Virginia and North Carolina provide a basis for comparison. The d13C and d15N values for the cave burials suggest a diet composed of primarily C4 plant proteins and some terrestrial animal proteins.

Genetic divergence and evolutionary times: calibrating a protein clock for South-European Stenasellus species (Crustacea, Isopoda), 1997, Argano Roberto, Sbordoni Marina Cobolli, Matthaeis Elvira De, Ketmaier Valerio
We studied genetic divergence in a group of exclusively stygobiont isopods of the family Stenasellidae. In particular, we assessed evolutionary relationships among several populations of Stenasellus racovitzai and Stenasellus virei. To place this study in a phylogenetic context. we used another species of Stenasellus, S. assorgiai, as an outgroup. S. racovitzai occurs in Corsica, Sardinia and in the fossil islands of the Tuscan Archipelago, while S. virei is a polytypic species widely distributed in the central France and Pyrenean area. This vicariant distribution is believed to be the result of the disjunction of the Sardinia-Corsica microplate from the Pyrenean region and its subsequent rotation. Since geological data provide time estimates for these events, we can use the genetic distance data to calibrate a molecular clock for this group of stygobiont isopods. The calibration of the molecular clock reveals a roughly linear relationship (r = 0.753) between the genetic distances and absolute divergence times, with a mean divergence rate (19.269 Myr/DNei,) different from those previously reported in the literature and provides an opportunity to shed some light on the evolutionary scenarios of other Stenasellus species.

Fluorescence wavelength and intensity variations of cave waters, 1999, Baker A, Genty D,
The fluorescence properties of groundwaters percolating into four cave systems have been monitored over the period 1997-1998. Fluorescence was excited between 220 and 400 nm and the emission measured from 300 to 500 nm using a fluorescence spectrophotometer. Three fluorescence centres were observed; one at the excitation-emission pair of 290-340:395-430 nm, (humic-like, probably fulvic acid), one at 265-280:300-370 nm (protein like) and a less defined region of high fluorescence at 230-280:310-420 nm (humic and/or protein like). The most consistent fluorescence intensity was observed in the excitation-emission pair of 290-340:395-430 nm, attributed to a fulvic acid source. Subtle differences (5%) in the fluorescence excitation and emission wavelength of this fluorescence peak in the groundwater were observed between the four sites, and the fluorescence intensity varied considerably ( x 60) between the four sites. Both the wavelength and the intensity variations in fluorescence are caused by the differences in the vegetation cover, soil type and humification. Data from the most intensely monitored site (Brown’s Folly Mine, England; 9 sample stations, 10-20 days frequency sampling) revealed no spatial variability in the 290-340:395-430 nm (fulvic acid) fluorescence; in contrast time-series analysis suggests that the seasonal variations do occur, with a decrease in the emission wavelength correlating with the first (autumn) peak in fluorescence intensity, and a decrease in the excitation wavelength correlating with a second (winter) fluorescence intensity peak. Results demonstrate the potential of utilising fluorescence wavelength variations in sourcing karst groundwaters, and as a possible palaeoenvironmental proxy of the overlying soil conditions if trapped within the cave speleothems

The presence of original organic matter and retention of an indigenous isotopic signal in fossils have been disputed for years. An experiment was conducted to evaluate the influence of diagenesis on bone-protein isotope values, analyses were conducted on Holocene and Pleistocene fossils from Natural Trap Cave (NTC), Wyoming. Modern cow, Bos taurus, bone was heated with and without excess water for up to 195 hours at 100{degrees}C in an inert atmosphere. Collagen and non-collagenous proteins (NCP) were extracted and analyzed isotopically. Under dry conditions, carbon and nitrogen isotope values change by less than 0.4{per thousand} during the 0 to 195 hour interval. In the presence of excess water, carbon and nitrogen isotope values change by no more than 1.0{per thousand} and 0.5{per thousand}, respectively, over 192 hours. The relative abundance of amino acids of collagen from heated bone differs by less than 10% from that of unheated collagen. Protein preservation is indicated by matrix-assisted laser desorption ionization mass spectrometry (MALDI-MS) that strongly indicates a portion of the original osteocalcin exists intact in samples heated for 195 hours. Good preservation of collagen in NTC fossils is implied by high collagen yields, C:N, and realistic trophic structures based on isotope values. Carbon and nitrogen isotope values of ancient collagen increase with trophic level, allow dietary assessments to be made, and differentiate between ruminant and non-ruminants. The results indicate that isotope values are resilient during simulated diagenesis and suggest that an indigenous isotopic signal can exist in well-preserved fossils such as those from NTC

Dosimetric and radiocarbon chronology of a pre-Wisconsinan mastodon fossil locality at East Milford, Nova Scotia, Canada, 2003, Godfreysmith D. I. , Grist A. M. , Stea R. R. ,
We report the final results of a multidisciplinary geochronological study of two subfossil mastodon remains and the sediments associated with them. from the East Milford, N.S. mastodon locality discovered in 1991. The mastodons, which were found in a karstic sinkhole system, are the latest and the most complete of several mastodon remains discovered in Nova Scotia, New Brunswick and Maine over the last 170 years. The unconsolidated sediments containing the adult specimen were dated using optical dating (IRSL); the mastodon dental enamel was dated using electron spin resonance (ESR); and bone collagen from both individuals plus associated wood were dated using radiocarbon (C-14) dating. Two samples of sediment adhering to the walls of the karstic cavity at the elevation corresponding to the location of the adult specimen yielded statistically indistinguishable IRSL ages of 127 13 (EMM1) and 143 16 ka (EMM2), with a weighted mean of 133 6 ka. This is consistent with pollen data which indicate an interglacial climate. On the basis of its stratigraphic position with respect to overlying till units, the locality is attributed to oxygen isotope stage 5. Three ESR dates on dental enamel (natural prompt, deproteinated prompt, and deproteinated delayed) yielded statistically indistinguishable ages whose weighted mean is 74.9 5.0 ka. This indicates consignment to the geologic record of the mastodon tooth during terminal oxygen isotope stage 5a, when the climatic cooling leading to the Wisconsinan glacial period had already begun. Fission-track analysis of the enamel and dentine cross sections showed minimal U uptake, resulting in no need for early, linear, or late uptake modelling. The direct ESR age indicates that the specimen either became mired in a cavity that was already at least partly infilled with older, probably waterlogged, sediments, or that the sediment's IRSL signal was not completely erased during deposition. The dosimetric ages are consistent with limiting radiocarbon ages on bone collagen (juvenile > 45,760 years BP; adult greater than or equal to 37,040 1730 BP) and wood (near the adult mastodon > 51,000 BP; organic-rich horizon above the karst surface > 50,000 BP). These results provide the first direct age on a mastodon fossil from Atlantic Canada, and the first numeric estimate to demonstrate that mammoths were present in this region during pre-Wisconsinan times, specifically during late oxygen isotope stage 5a. Variability in terrestrial and microbial contributions to dissolved organic matter fluorescence in the Edwards Aquifer, Central Texas, 2009, Birdwell J. E. And Engel A. S.
Most cave and karst ecosystems are believed to be dependent on an influx of allochthonous organic carbon. Although microbes are largely responsible for the fate of dissolved organic matter (DOM) in karst, the role of microbes in chemosynthetic (autochthonous) production and processing of DOM has received limited attention. Chromophoric dissolved organic matter (CDOM) is the fraction of DOM that absorbs ultraviolet and visible light, and differences in the fluorescence spectral characteristics of humic-like (terrigenous) and protein-like (microbially-derived) CDOM allow for tracing the relative contributions of allochthonous or autochthonous carbon sources, respectively, in water. We investigated CDOM in karst-aquifer well and spring waters along the fresh- to saline-water transition zone of the Edwards Aquifer, Central Texas, over a four year period. The groundwater fluorescence spectral characteristics were distinct from those generally observed in surface waters and soil porewaters. The dominant source of organic carbon in the aquifer waters may be a product of chemolithoautotrophic primary production occurring in situ. It is possible that the absence of a strong terrestrial CDOM signature may be due to filtering effects in the epikarst or rapid utilization by heterotrophs in the aquifer. Our results indicate that intense recharge following periods of drought may influence the intensity of microbial activity, either due to an influx of DOM or nutrients from the surface that was not quantified by our analyses or because of increased in situ autotrophic activity, or both. The variable contributions of allochthonous and autochthonous DOM during and after recharge events call into question whether karst aquifer ecosystems are necessarily dependent on allochthonous organic matter.

Diatom flora in subterranean ecosystems: a review., 2014,

In scarcity of light and primary producers, subterranean ecosystems are generally extremely oligotrophic habitats, receiving poor supplies of degradable organic matter from the surface. Human direct impacts on cave ecosystems mainly derive from intensive tourism and recreational caving, causing important alterations to the whole subterranean environment. In particular, artificial lighting systems in show caves support the growth of autotrophic organisms (the so-called lampenflora), mainly composed of cyanobacteria, diatoms, chlorophytes, mosses and ferns producing exocellular polymeric substances (EPSs) made of polysaccharides, proteins, lipids and nucleic acids. This anionic EPSs matrix mediates to the intercellular communications and participates to the chemical exchanges with the substratum, inducing the adsorption of cations and dissolved organic molecules from the cave formations (speleothems). Coupled with the metabolic activities of heterotrophic microorganisms colonising such layer (biofilm), this phenomenon may lead to the corrosion of the mineral surfaces. In this review, we investigate the formation of biofilms, especially of diatom-dominated ones, as a consequence of artificial lighting and its impacts on speleothems. Whenever light reaches the subterranean habitat (both artificially and naturally) a relative high number of species of diatoms may indeed colonise it. Cave entrances, artificially illuminated walls and speleothems inside the cave are generally the preferred substrates. This review focuses on the diatom flora colonising subterranean habitats, summarizing the information contained in all the scientific papers published from 1900 up to date. In this review we provide a complete checklist of the diatom taxa recorded in subterranean habitats, including a total of 363 taxa, belonging to 82 genera. The most frequent and abundant species recorded in caves and other low light subterranean habitats are generally aerophilic and cosmopolitan. These are, in order of frequency: Hantzschia amphioxys, Diadesmis contenta, Orthoseira roeseana, Luticola nivalis, Pinnularia borealis, Diadesmis biceps and Luticola mutica. Due to the peculiarity of the subterranean habitats, the record of rare or new species is relatively common. The most important environmental factors driving species composition and morphological modifications observed in subterranean populations are analysed throughout the text and tables. In addition, suggestions to prevent and remove the corrosive biofilms in view of an environmentally sustainable cave management are discussed.
